Given that we have some preliminary data on TCRs being MC38B reactive, we would like to explore their respective targets. For this, we need to

  1. get the cell line sequenced against our B6 host (as normal) (Get MC38B cell line RNA- and WEX-sequenced #5)
  2. knock out either H2Kb or H2Db and test these variants against our TCRs to see if either of them will eliminate the cytotoxicity (Knock H2Kb and H2Db out from MC38Bs #6)
  3. try previously described MC38 tetramers on the cells (Try to see if our MC38B TCRs can recognize previously described epitope candidates #7)
  4. adopt a DNA-library-based presentation system (e.g. PresentER) to screen for hundreds of candidates

A few months ago I had to review this topic and put my notes up on Discourse. I need to update that topic with the new work that has landed in the past few months but I'd be happy to share what I've learned if we get past step 3.
