Hello! Thank you for the great program.
I was just wondering if there is a difference between 5’ 10x data and 3’ data on velocity analysis?
I'm using 10x Immune Profiling gene expression kit which sequences RNA from 5', and the average spliced/unspliced ratio is 26:74 (and all clusters are pretty much similar in ratio).
This is opposite to what I would in most examples. Would the 5' technology affect the results? and if so, how?
If not, would this be due to the type of samples (PBMC) that we used?
